Description
The TP53 gene encodes the tumor protein p53, a tumor suppressor protein containing transcriptional activation, DNA binding, and oligomerization domains. The encoded protein responds to diverse cellular stresses to regulate expression of target genes, thereby inducing cell cycle arrest, apoptosis, senescence, DNA repair, or changes in metabolism. In unstressed cells, p53 is kept inactive essentially through the actions of the ubiquitin ligase MDM2 (OMIM: 164785), which inhibits p53 transcriptional activity and ubiquitinates p53 to promote its degradation.Mutations in this gene are associated with a variety of human cancers, including hereditary cancers such as Li-Fraumeni syndrome (Toledo and Wahl, 2006; Bourdon, 2007; Vousden and Lane, 2007).
